Starch (Acid hydrolysis not necessary)

Expanding Blue (acid hydrolyzes cellulose to hydrocellulose which reacts with IKI)

1.  Place sections in drop of IKI solution on slide and cover with coverslip.
 
2.  Allow drop of 65% Sulphuric Acid to diffuse underneath one side of coverslip while watching for swelling of cellulose.
1.  IKI solution = 1 g I and 3 g KI in 300 ml of dH2O.

2. 65% aqueous Sulphuric Acid 

IMPORTANT NOTES:
  1. ACID SHOULD BE INTRODUCED ON LARGEST FREE SURFACE OF MICROSCOPE SLIDE TO PROTECT THE MICROSCOPE!!!
  2. GREAT CARE SHOULD BE EXERCISED TO AVOID CONTACT OF ACID WITH MICROSCOPE!!!
